MiniLeafCrusher

The hand-held MiniLeafCrusherTM is primarily designed for, but not restricted to, use in the field or locations distant from a diagnostic laboratory.  This modified c-clamp is used in combination with a BioSpec DNA** Sample CardTM containing a small sheet of WhatmanTM cellulose filter paper.  Plant cells crushed by the clamp are disrupted and the released cell extract is immediately captured and stabilized on the Sample Card.  Added preservatives or cooling are not required during transit of the card(s) to the lab for subsequent analysis.

** Plant leaf biochemicals, such as RNA and proteins, can also be collected and preserved with the MiniLeafCrusher and its Sample Card.  The crushing tool performs best with plant leaf tissue.  Animal tissue cell walls are too soft and flexible to be completely "crushed" in the jaws of the MiniLeafCrusher.  The Sample Card can also be used alone to stabilize and transport a drop-sized fluid suspension of tissue homogenate prepared by other means (see "Description" below), bodily fluids (blood or urine) or liquid suspensions of biological/environmental material (feces, lake mud, processed foods).

HOW IT WORKS:  A plant leaf is inserted in the BioSpec DNA* Sample Card and the loaded Sample Card is magnetically docked between the jaws of the MiniLeafCrusher.  Manually tightening the Crusher generates a very high crushing force on a thumb-tack-sized area of the plant leaf.  The crushed plant cells are totally disrupted and the resultant cell lysate is captured on a layer of cellulose filter paper located in the Sample Card.  The Sample Card is folded closed and packaged in a provided zip-lock polybag containing a pouch of desiccant. The sample extract is preserved in a dried and stabilized state ready for later delivery to the laboratory for analysis.

On delivery to the lab, the sample spot can be cut out of the Sample Card's filter paper sheet in the shape of a dipstick or punched out as a 1/4" disk, and then eluted in an appropriate extraction medium.  For details on the rapid and efficient recovery and PCR amplification of plant leaf DNA from the filter paper, see Zou et.al., PLoS Bio 15 (11), e2003916. https://doi.org/10.1371/journal.pbio.2003916.

Other "field-compatible" homogenizers suitable for the isolation of nucleic acids and proteins from plant leaf material available from BioSpec Products (see Spiralpestle and Microtube Homogenizer and Portable SoniBeast-Jr.).  While not as simple to use as the MiniLeafCrusher, these tools have the advantage of being applicable to animal tissue as well as almost all plant tissues, including seeds and roots.
